In this paper, we propose a general copula approach to accommodate non-normal continuous mixing distributions in multinomial probit models. In particular, we specify a multivariate mixing distribution that allows different marginal continuous parametric distributions for different coefficients. A new hybrid estimation technique is proposed to estimate the model, which combines the advantageous features of each of the maximum simulated likelihood inference technique and Bhat’s maximum approximate composite marginal likelihood inference approach. Flavin‐containing monooxygenases (FMOs), traditionally known for detoxifying xenobiotics, are now recognized for their involvement in endogenous metabolism. We recently discovered that an isoform of FMO, fmo‐2 in Caenorhabditis elegans, alters endogenous metabolism to impact longevity and stress tolerance. Increased expression of fmo‐2 in C. elegans modifies the flux through the key pathway known as One Carbon Metabolism (OCM). This modified flux results in a decrease in the ratio of S‐adenosyl‐methionine (SAM) to S‐adenosyl‐homocysteine (SAH), consequently diminishing methylation capacity. Here we discuss (...) how FMO‐2‐mediated formate production during tryptophan metabolism may serve as a trigger for changing the flux in OCM. We suggest formate bridges tryptophan and OCM, altering metabolic flux away from methylation during fmo‐2 overexpression. Additionally, we highlight how these metabolic results intersect with the mTOR and AMPK pathways, in addition to mitochondrial metabolism. In conclusion, the goal of this essay is to bring attention to the central role of FMO enzymes but lack of understanding of their mechanisms. We justify a call for a deeper understanding of FMO enzyme's role in metabolic rewiring through tryptophan/formate or other yet unidentified substrates. Additionally, we emphasize the identification of novel drugs and microbes to induce FMO activity and extend lifespan. (shrink)

Human-eyelid conditioning was the principal source of information on Pavlovian conditioning, especially human, in the 1950s and 1960s, but it suffered a sharp decline in productivity, beginning in the late 1960s. The present article treats the decline as a case study with potential implications concerning the survival contingencies of research specialties. We make use of questionnaire data from eyelid-conditioning researchers and examine a variety of publication, topic-of-investigation, and institutional data to identify the major factors in the decline of human-eyelid conditioning.

It is suggested that the control-system theory of Domjan et al. restates in engineering vocabulary the primary thesis of law-of-effect theories: namely, that classical-conditioning arrangements may involve CR-contingent reinforcement. The research cited by Domjan et al. is relevant to the idea that classical conditioning is an adaptive process, but is irrelevant to their control-system theory.

In two chapters of Enn. II.9 [33], Plotinus discusses the Gnostic idea that the creation of the world is due to the ‘decline’ of a principle that he variously calls Soul or Sophia. The identity of Plotinus' Gnostics is notoriously difficult to establish with any degree of precision; I can only note here that the idea of Sophia's ‘decline’ features in a number of extant Gnostic texts, such as those from Nag Hammadi and the Berlin Codex, as a recent survey (...) of the evidence by Poirier has demonstrated. The ArgumentMichael Polanyi is placed in the ongoing Enlightenment-reform tradition as one of the first twentieth-century scientists to propose a program to correct the gravest internal conflict of the modern Enlightenment project of radical criticism: scientific detachment and moral nihilism in conflict with humanist values. He held that radical criticism leads not to truth but to destructive doubt. Only the inclusion of the “personal element,” the judicial attitude of reasonable doubt and the acknowledgment of belief in the regulative principle of (...) truth can overcome this end. Freedom — to reason, to criticize the method of doubt, and freedom of scientific thinking — is a precondition to reach truth. His epistemology of nonsubjective personal knowledge has redefined scientific knowing and objectivity as grounded in tacit knowing. Debate is continuing on objectivity and freedom versus the social duty of science. Polanyi's Hungarian correspondence highlights his commitment to truth and the contemporary cultural-political situation of Jewish intellectuals. (shrink)

Many religious people are alarmed about features of the current age - violence in the media, a pervasive hedonism, a marginalization of religion, and widespread abortion. These concerns influence politics, but just as there should be a separation between church and state, so should there be a balance between religious commitments and secular arguments calling for social reforms. The discovery of selective associability of cues in classical conditioning has often been treated as an embarrassment to Pavlov, because he has been represented as a proponent of the "equivalence of associability of cues." According to that doctrine, except for the influence of differences in stimulus intensity, all environmental stimuli are equally susceptible to becoming conditioned stimuli if they are arranged in a suitable time-relation to any effective unconditioned stimulus . The current paper asks whether Pavlov explicitly made such a (...) claim and, if not, whether he could have endorsed equivalence of associability. Scientific controversy, the role that "the classics" play in scientific specialties, and the emblematic standing of the founding figures of a discipline or specialty constitute a framework for discussion of Pavlov's stand on the equivalence of associability. In Ukrainian church life, the influence of the Byzantine Empire, which has existed for over eleven centuries, holds a special place. This unique Greek superpower became the first independent state where faith in Jesus Christ became part of the entire state complex. It was this faith that united Byzantium with the Ecumenical Church, whose center of history was rooted in Rome.

Following promulgation of the Nuremberg code in 1947, the ethics of research on human subjects has been a challenging and often contentious topic of debate. Escalation in the use of research participants in low-income countries over recent decades , has intensified the debate on the ethics of international research and led to increasing attention both to exploitation of vulnerable subjects and to considerations of how the 10:90 gap in health and medical research could be narrowed. In 2000, prompted by the (...) discussions over several years that led to the US NIH launching a capacity building programme on research ethics for members of research ethics committees in developing countries, we advanced a ‘new look’ for the ethics of international research.1 Since then progress has been made on several fronts.First, our ideas—considered somewhat radical and impractical at the time—have been provocatively addressed by scholars who have either contested them or advanced similar conceptions of what obligations international researchers have to research participants and communities in low income countries before, during and after clinical trials. Second, those researchers who have been sympathetic to our ideas have either endeavoured to put these into practice or have investigated the feasibility of doing so. Third, the intractability of the 10/90 gap and the escalation of interest in global health have sensitised many to the need to amplify the uptake of these ideas in practice.Here, we briefly review the conceptual and practical developments in international research ethics. We present a model of steady state solute and water reabsorption along the rat proximal tubule. Major co-and counter-transport systems in the apical and basolateral cell membranes are described using kinetic descriptions based on data from the flows and solute concentrations along the length of the proximal tubule as a function of filtration rate and peritubular solute concentrations. We show that for many aspects of proximal tubule transport physiology this kinetics-based model is an adequate representation of the mammalian proximal tubule.
